Abstract: In this study, three monoclonal antibodies (MAbs) against Salmonella were produced from mice immunized with a combination of heat-killed and formalin-fixed of Salmonella Enteritidis alone or a mixture of four Salmonella serovars (S. Enteritidis, S. Typhimurium, S. Hadar and S. Virchow). The first MAb, SE-13D is IgM, which bound specifically and only to S. Enteritidis. The second MAb Sal-06G is IgG2a, and bound to the Salmonella serogroups B, D, E, I and some serovars in group C (S. Winston and S. Bovismorbificans). The third, MAb Sal-08G is IgG1, bound to most Salmonella serovars but cross-reacted to Shigella dysenteriae and Escherichia coli. Using a dot-blot assay, the detection sensitivity limits of MAbs ranged from 2.5 x 106 - 107 cfu/mL. However, the detection of these MAbs could be improved to 10 or 1 cfu/mL after pre-enriching the bacterial sample in tryptic soy broth (TSB) or chicken homogenate in TSB for 6 or 12 h, respectively. Therefore, these MAbs can be used as simple, rapid, specific, and effective immunological tool for direct detection and differentiation of S. Enteritidis from an assortment of Salmonella serovars from other bacteria in complex samples including poultry products, foods, and medical samples without the need for bacterial isolation and the conventional characterizations which is required by series of biochemical tests.
งานวิจัยในครั้งนี้ได้ทำการผลิตโมโนโคลนอลแอนติบอดีต่อเชื้อ Salmonella โดยการปลูกภูมิคุ้มกันในหนูด้วยเชื้อ Salmonella Enteritidis เท่านั้นหรือเชื้อ Salmonella 4 ซีโรวาร์ ผสมกัน (S. Enteritidis, S. Typhimurium, S. Hadar และ S.  Virchow) ในรูปแบบที่ทำให้ตายด้วยความร้อนผสมกับรูปแบบที่คงสภาพด้วยฟอร์มาลิน สามารถผลิตโมโนโคลนอลแอนติบอดีได้ 3 ชนิด ซึ่งมีความจำเพาะที่แตกต่างกัน โดยโมโนโคลนอลแอนติบอดีชนิดที่ 1 (SE-13D) เป็น IgM มีความจำเพาะต่อเชื้อ S. Enteritidis เท่านั้น โมโนโคลนอลแอนติบอดีชนิดที่ 2 (Sal-06G) เป็น IgG2a สามารถจับกับเชื้อ Salmonella serogroups B, D, E, I และบางซีโรวาร์ในกลุ่ม C (S. Winston และ S. Bovismorbificans) โมโนโคลนอลแอนติบอดีชนิดที่ 3 (Sal-08G) เป็นชนิด IgG1 สามารถจับกับเชื้อ Salmonella ส่วนใหญ่ แต่มีปฏิกิริยาข้ามกับเชื้อ Shigella dysenteriae และ Escherichia coli โดยโมโนโคลนอลแอนติบอดีทั้ง 3 ชนิดที่ผลิตได้ มีความไวในการตรวจอยู่ในช่วง 2.5 x 106 - 107 cfu/mL เมื่อทดสอบโดยวิธี dot blotting อย่างไรก็ตามสามารถเพิ่มความไวในการตรวจด้วยการเพิ่มจำนวนเชื้อ  S. Enteritidis ในอาหารเลี้ยงเชื้อ TSB เป็นเวลา 6-12 ชั่วโมง ทำให้สามารถตรวจพบแบคทีเรียในตัวอย่างเพียง 10 - 1 cfu/mL ตามลำดับ และเมื่อทำการทดสอบบ่มเชื้อ S. Enteritidis กับตัวอย่างเนื้อไก่บดในอาหารเลี้ยงเชื้อ tryptic soy broth (TSB) ก็ยังคงให้ผลการทดสอบที่คล้ายคลึงกัน ดังนั้นโมโนโคลนอลแอนติบอดีที่ผลิตได้ในครั้งนี้ สามารถนำมาใช้ในการพัฒนาเทคนิคการตรวจวิเคราะห์เพื่อจำแนกเชื้อ S. Enteritidis ออกจาก Salmonella ซีโรวาร์อื่น และแบคทีเรียต่างชนิด โดยวิธี dot blotting ซึ่งเป็นการตรวจวิเคราะห์ที่มีความไวและความแม่นยำสูง ใช้เวลาในการตรวจน้อย ไม่จำเป็นต้องทำการแยกเชื้อและทดสอบทางชีวเคมี  ทำให้สามารถตรวจสอบการปนเปื้อนของ เชื้อSalmonella ได้อย่างรวดเร็วจึงช่วยป้องกันและเฝ้าระวังการแพร่ระบาดของเชื้อ S. Enteritidis ที่เป็นสาเหตุหลักของโรค salmonellosis
